The CryoEM Method MicroED as a Powerful Tool for Small Molecule Structure Determination … reports on use of the cryo-EM method microcrystal electron diffraction (MicroED) to provide routine and unambiguous structural determination of small organic molecules. In 2013 the cryoelectron microscopy (cryo-EM) technique called microcrystal electron diffraction (MicroED) was first established and reported upon. MicroED data are collected as movies while the stage of the cryo-EM is continuously rotated. This produces a series of high-resolution diffraction patterns that can be processed to produce high-resolution structures directly from microcrystals. Since that time, data collection and analysis schemes have been fine-tuned, and structures for over 40 different proteins, oligopeptides and organic molecules and been determined. Now, starting with simple powders, with minimal sample preparation, the authors report on collecting high quality MicroED data from nanocrystals (~100x100x100 nm, ~10–15g) resulting in atomic resolution (<1 Å) crystal structures in minutes.
